The aft tank being lower than the forward tank & the fwd tank over filling with both tank valves open, that is very informative!

I have used Teflon tape on threads of both fwd & aft tanks threaded access port (clean out plug) & inadvertently overfilled both tanks, the Teflon tape sealed & the only overflow was through the tank vents.
My initial response to Dianne's request:
If I discovered water in the aft stateroom under the seat after filling the forward tank, would I consider that the tank overflowed and the fresh water drained aft to the stateroom bilge?
If the forward water tank overfilled I would expect the excess water would spill from the vent into the anchor locker and drain overboard.
If water overflowed from the threaded cap on the top of the forward water tank there would be evidence of water spill down the tank flowing after under the bilge liner to the lowest part of the boat, the bilge alongside the holding tank.
There would have to be water in this starboard bilge area between the holding tank and the vertical panel. Also in the port bilge under the galley drawer cabinet. The water level would have to be high here in order to reach under the aft stateroom seat.
If the forward tank had a leak, the water level would drop and a tank leak would evident. The water would drain to the lower bilge.
However I don’t think that water over flow from the forward tank would drain aft to the stateroom bilge. First I would check by the holding tank. And I would hand pump the water out.
I would also check the aft water tank water level for leaks. And the water tube fittings for leaks.
Maybe pour blue food color into one tank at a time to see if blue water turns up in the bilge.
Another place I would look is behind the aft state room drawers. The wood drawer assembly is easily removed by 5 wood screws. Here in the bilge are the seacocks and hoses for the Head sink drain, shower sump & toilet raw water suction.
Also the hot & cold fresh water supply to sink in the Head. Possible a water leak here would migrate to the area under the aft stateroom seat. Look at the anti-siphon tube too.
I cannot remove the floor covering on my galley sole to locate the access opening shown on the drawing. So it is possible there is water in the bilge area under the sole. For this bilge area I look under both the galley drawer cabinet and next to the holding tank for water.
Another unlikely fresh water source would be the 15mm tubing from the aft water tank going forward through the bilge, alongside the diesel bilge, under the galley sole and turning upwards to the water pump and galley sink. A fresh water leak from the tubing or fitting? Blue food color in the water tank might reveal a leak.
Finally I might suspect the water dripping from the packing gland that is not draining through the center line tube going forward under the diesel bilge to drain in the ‘bilge pan’ bilge pump location. If this tube is blocked up the water level would rise and drain into the holes cut for the 15mm tubing from the aft water tank in to the area under the aft stateroom seat.
Maybe sprinkle green Comet powder around under the seat in the unlikely event the water is leaking from above, a deck fitting or window. The water would leave a trail in the green powder.
I hope my comments initiate a conversation, this has had to have happen on another C320.
